Comfort II new boards has Kate29 as vocabulary. And kate29.bin can be downloaded from www.cytech.biz.
Differently from kate28 is is just one file, not full package. How to modify this vocabulary without having kate29.ini and other files. Do I am mixing something?

kate29.bin is the binary file which is transferred to Comfort using Comfigurator > Transfer > Vocabulary Upgrade. Are you trying to translate Comfort into another language?
For this you need the VoiceProc and VoicePlay software which you can download from www.cytech.biz/software.html
Instructions are in the voiceproc.pdf document download from here
The configuration files for kate29 are attached, for the Engish language

I will add some missting words. There is an Idea to make a translation to another language but at first we need to learn how all process works before starting this translating/recording/editing/updating job
